CJC-1295 without Drug Affinity Complex (also known as modified GRF 1-29 or mod-GRF) is a synthetic 29-amino acid analog of growth hormone-releasing hormone (GHRH) with four amino acid substitutions that confer resistance to DPP-IV enzymatic degradation. Without the DAC (maleimidopropionic acid) conjugation that enables irreversible albumin binding, this form has a shorter half-life but produces more physiologic pulsatile GH release patterns, making it preferred for research protocols simulating natural GH secretion.

Mechanism of Action
CJC-1295 (no DAC) binds the GHRH receptor on anterior pituitary somatotrophs, activating Gs-adenylyl cyclase-cAMP-PKA signaling to stimulate GH synthesis and secretion. The four amino acid modifications (Ala at positions 2, 8, 15; Leu at position 27) prevent DPP-IV cleavage between His-Ala at position 1-2, extending activity from ~7 minutes (native GHRH) to ~30 minutes. Without DAC, it does not sustain albumin binding, producing acute GH pulses rather than sustained GH elevation.
