Mechanism of Action
MT-2 binds melanocortin receptors 1-5 with varying affinity. MC1R activation on melanocytes upregulates tyrosinase and melanin synthesis via cAMP/PKA signaling. MC4R activation in hypothalamic nuclei modulates MC4R arousal-circuit signaling and feeding-circuit pathways studied in preclinical models. MC3R engagement influences energy-metabolism signaling and lipid partitioning. The cyclic structure of MT-2 (Ac-Nle-c[Asp-His-D-Phe-Arg-Trp-Lys]-NH₂) provides enhanced receptor binding affinity and enzymatic resistance over linear α-MSH.
