SLU-PP-332 is a small-molecule agonist of estrogen-related receptor (ERR) alpha, beta, and gamma — orphan nuclear receptors that regulate mitochondrial biogenesis, oxidative metabolism, and exercise adaptation gene programs. Developed at Saint Louis University, SLU-PP-332 has been described in mouse studies as an 'exercise mimetic' because it induced an ERRα-dependent acute aerobic-exercise gene program in skeletal muscle. Mechanism of Action
SLU-PP-332 is a pan-ERR agonist that targets ERRα, ERRβ, and ERRγ, with the highest potency at ERRα. In a skeletal muscle cell line it increased mitochondrial function and cellular respiration, and in mice it increased type IIa oxidative skeletal muscle fibers. In a treadmill run-to-exhaustion study in sedentary male mice dosed 50 mg/kg intraperitoneally twice daily for about a week, treated animals ran approximately 70 percent longer and approximately 45 percent further than vehicle controls, and the effect was abolished by muscle-specific ERRα deletion.
