SLU-PP-332 is a small-molecule agonist of estrogen-related receptor (ERR) alpha, beta, and gamma — orphan nuclear receptors that regulate mitochondrial biogenesis, oxidative metabolism, and exercise adaptation gene programs. Developed at Saint Louis University, SLU-PP-332 has been described as an 'exercise mimetic' because it activates transcriptional programs normally induced by endurance exercise, including fatty acid oxidation, mitochondrial biogenesis, and muscle fiber-type switching, without physical activity.

Mechanism of Action
SLU-PP-332 binds to ERRα, ERRβ, and ERRγ, stabilizing the active conformation and promoting coactivator recruitment (PGC-1α, SRC family). This activates transcription of genes governing mitochondrial biogenesis (TFAM, NRF1/2), oxidative phosphorylation (cytochrome c, ATP synthase subunits), fatty acid β-oxidation (CPT1, ACADM, HADH), and slow-twitch muscle fiber programs (MYH7, troponin I slow). In murine models, SLU-PP-332 increased treadmill endurance by approximately 50% and enhanced resistance to muscle fatigue.
