SNAP-8 (acetyl octapeptide-3) is a synthetic octapeptide that mimics the N-terminal domain of SNAP-25 (synaptosomal-associated protein of 25 kDa), one of the three SNARE complex proteins required for vesicular neurotransmitter release. It is an extension of the hexapeptide acetyl hexapeptide-3 (Argireline) with two additional amino acids, providing enhanced potency. SNAP-8 is primarily investigated in dermatological research as a topical neuropeptide for expression line modulation.

Mechanism of Action

SNAP-8 competitively inhibits SNARE complex assembly by mimicking the SNAP-25 protein, thereby attenuating vesicular acetylcholine release at the neuromuscular junction of facial muscles. By reducing the efficiency of synaptic vesicle docking and fusion, SNAP-8 decreases the intensity of muscle contraction underlying expression lines. Unlike botulinum toxin (which cleaves SNARE proteins proteolytically and irreversibly), SNAP-8 acts as a reversible competitive antagonist.
