Human chorionic gonadotropin (HCG) is a glycoprotein hormone consisting of an alpha subunit (common to LH, FSH, TSH) and a unique beta subunit that confers biological specificity. HCG mimics luteinizing hormone (LH) activity by binding the LH/CG receptor on Leydig cells and ovarian theca cells. It is a non-covalent heterodimer of a 92-residue alpha subunit (UniProt P01215, shared with LH, FSH and TSH) and a 145-residue beta subunit (UniProt P0DN86) that confers its identity; no single molecular formula can be assigned, because eight glycosylation sites carry carbohydrate that varies between preparations (~30% of mass; unglycosylated backbone ~25,700 Da). It has a significantly longer half-life than LH due to its greater degree of glycosylation, making it a durable LH-receptor agonist for research applications requiring sustained Leydig cell or theca cell stimulation. Mechanism of Action

HCG binds the LH/CG receptor (LHCGR), a Gs-coupled GPCR on testicular Leydig cells, activating cAMP/PKA signaling that stimulates steroidogenic acute regulatory protein (StAR) and cholesterol side-chain cleavage enzyme (CYP11A1), thereby driving de novo testosterone biosynthesis. In ovarian tissue, HCG triggers final oocyte maturation, ovulation, and corpus luteum formation. HCG also maintains intratesticular testosterone concentrations necessary for spermatogenesis, independently of pituitary LH.
