Semax is a synthetic heptapeptide (Met-Glu-His-Phe-Pro-Gly-Pro) derived from the N-terminal region of adrenocorticotropic hormone: it retains the ACTH(4-7) fragment Met-Glu-His-Phe and substitutes a Pro-Gly-Pro tripeptide for residues 8-10 (Arg-Trp-Gly). Developed at the Institute of Molecular Genetics of the Russian Academy of Sciences, Semax is classified as a nootropic and neuroprotective agent in Russian pharmacopoeia. It has been studied extensively in cognition and memory models, post-ischemic neuroprotection models, and for modulation of neurotrophic factor expression. Mechanism of Action
In rats, Dolotov et al. reported increased BDNF and TrkB expression in the hippocampus after a single intranasal exposure to Semax. Agapova et al. reported increased NGF gene expression in rat hippocampus but decreased NGF gene expression in rat frontal cortex after a single intranasal exposure. It modulates the melanocortin system via MC4R interaction without significant adrenal steroidogenic activity in the studied dose ranges. Additional mechanisms include modulation of serotonergic and dopaminergic neurotransmission.
