Semax is a synthetic heptapeptide (Met-Glu-His-Phe-Pro-Gly-Pro) derived from the N-terminal fragment of adrenocorticotropic hormone (ACTH 4-10), with a C-terminal Pro-Gly-Pro tripeptide extension conferring enhanced metabolic stability. Developed at the Institute of Molecular Genetics of the Russian Academy of Sciences, Semax is classified as a nootropic and neuroprotective agent in Russian pharmacopoeia. Mechanism of Action

Semax upregulates brain-derived neurotrophic factor (BDNF), nerve growth factor (NGF), and TrkB receptor expression in hippocampal and cortical regions. It modulates the melanocortin system via MC4R interaction without significant adrenal steroidogenic activity at nootropic doses. Additional mechanisms include attenuation of nitric oxide synthase activity in ischemic tissue, modulation of serotonergic and dopaminergic neurotransmission, and enhancement of cholinergic signaling relevant to memory consolidation.
