Mechanism of Action
Tesamorelin binds the GHRH receptor (GHRH-R) on pituitary somatotrophs, activating Gs-coupled cAMP/PKA signaling that stimulates GH gene transcription, GH synthesis, and GH secretory granule exocytosis. Unlike exogenous GH, tesamorelin preserves the pulsatile GH secretion pattern and maintains feedback sensitivity to somatostatin and IGF-1. In research models this pulsatile GH release is studied for visceral-adipose lipolysis signaling without the supraphysiologic IGF-1 elevation associated with exogenous GH.
