Mechanism of Action
Retatrutide is a triple agonist at GLP-1R, GIPR and the glucagon receptor (GCGR). Li et al. (2024) determined cryo-EM structures of retatrutide bound to each receptor in complex with Gs and examined receptor mutations using cAMP signaling assays. The study identifies shared and receptor-specific peptide contacts in these experimental systems.
